چکیده
This research provides data concerning the biochemical (ionic and organic) composition of sperm of the mirror carp (Cyprinus carpio) and its relationship to spermatozoa motility. Seminal plasma contained 67.12±1.06 mM/l Na+, 105.1±2.24 mM/l K+, 7.85±0.67 mg/dl Ca2+, 2.61±0.11 mEq/l Mg2+, 0.14±0.002 g/dl total protein, 10.3±1.01 mg/dl triglyceride, 6.83±0.72 mg/dl cholesterol, and 54.72±3.49 mg/dl urea. A positive relationship (p<0.05) was determined between Na+ and motility (r = 0.522). On the other hand, Ca2+, K+, and Mg2+ ions negatively correlated (p>0.05) with motility (r = -0.565, r = -0.160, and r = -0.184, respectively). Spermatozoa motility correlated negatively (p>0.05) with protein (r = -0.233), triglyceride (r = -0.348), and urea (r = -0.331) but positively with cholesterol (r = 0.012). This information will help to develop cryopreservation procedures, to meet species-specific extender requirements, and to optimize artificial fertilization procedures in mirror carp. Introduction Gamete management is key to achieving high fertilization success in fish farms (Billard et al. 1995). Studies on sperm physiology can provide basic knowledge for managing fertilization (Alavi and Cosson, 2005). For controlled and successful production in aquaculture systems, it is necessary to have adequate knowledge of the physical and chemical characteristics of the milt to determine the reproductive ability of cultivated fish. The ionic and organic constituents of the seminal fluid can indicate fish fertilization capacity (Ciereszko et al., 2000). * Corresponding author. E-mail: [email protected]

Effects of osmolality and potassium on motility of spermatozoa from freshwater cyprinid fishes.
Spermatozoa of freshwater Cyprinidae (goldfish, carp, crucian carp and dace) remained immotile when the semen was diluted in solutions of NaCl, KCl, mannitol or glucose iso-osmolar to the seminal plasma (300 mosmol kg-1). The spermatozoa became motile in media containing these solutes if the osmolality was lower than that of the seminal plasma, suggesting that motility is suppressed by the osmo...
متن کاملPreliminary studies on the cryopreservation of spermatozoa in the fresh water fish common carp (Cyprinus carpio L.)
Common carp [Cyprinus carpio (C. carpio)] is an economically important fish species cultured mostly in Asia and Europe. Global production of farmed common carp was about 6.14% (3 172 488 tonnes) of the total world aquaculture production[1]. Artificial insemination can be applied to increase the production to meet this demand due to exploitation of natural stocks. Cryopreservation is a valuable ...
متن کاملSperm characteristics in Grass carp, Ctenopharyngodon idella effect of ions on spermatozoa motility and fertilization capacity
In this study, sperm characteristics (motility parameters and ionic composition of seminal plasma) and effect of ions on motility (duration of motility of sperm and percentage of motile spermatozoa) and fertilization capacity (fertilization rate, hatching rate, survival rate and larvae length) were investigated in Ctenopharyngdon idella. The longest duration of motility obtained in solution con...
متن کاملEffects of different surfactants on motility and DNA integrity of brown trout (Salmo trutta fario) and common carp (Cyprinus carpio) spermatozoa.
In this study we evaluated effects of surfactants on motility parameters and DNA integrity of spermatozoa of freshwater teleost fish. Common carp (Cyprinus carpio) and brown trout (Salmo trutta fario) spermatozoa were exposed to either sodium dodecyl sulphate (SDS, anionic surfactant) or octoxynol 9 ( Triton X-100, nonionic surfactant). Both surfactants added at activation caused a decrease in ...
متن کاملProteomic characterization of fresh spermatozoa and supernatant after cryopreservation in relation to freezability of carp (Cyprinus carpio L) semen
Our recent studies suggested that the freezability of carp semen is related to seminal plasma protein profiles. Here, we aimed to compare the spermatozoa proteomes of good (GF) and poor (PF) freezability semen of carp. To achieve this, we used two-dimensional difference in gel electrophoresis followed by MALDI-TOF/TOF mass spectrometry. The semen was classified as GF or PF based on sperm motili...
